Macbeth got his first dose of ambition when considering the murder of Duncan. After he did murder Duncan, his ambition sky-rocketed, and he was able to kill anyone who stood in his way of ruling Scotland, including Banquo, his best friend.
Lady Macbeth seems to always be full ambition, so much so that she aided in the murder of King Duncan. In fact, she convinced Macbeth to kill Duncan. Without her in the play, Macbeth may have never killed Duncan. Before he kills Duncan, Macbeth's ambition appears to be fueled by Lady Macbeth.
